• 荔枝(Litchi chinesis Sonn.)果实酸性转化酶LcSAI 生物信息学和表达分析

    Abstract:酸性转化酶作为蔗糖代谢中的关键酶,在还原糖积累型荔枝中表达及酶活性显著高于蔗糖积累型荔枝。为了全面了解酸性转化酶的生物学特征,本研究通过运用生物信息学方法对荔枝果实酸性转化酶LcSAI 基本的理化性质、蛋白二级结构、亲水性/疏水性、跨膜结构域、信号肽、磷酸化位点、保守结构域、系统进化进行系统的分析;利用qRT-PCR 技术对LcSAI 在妃子笑不同组织和果实不同发育阶段进行表达分析。结果表明:荔枝果实酸性转化酶为定位于液泡的亲水性不稳定蛋白,无信号肽,其蛋白的二级结构主要有无规则卷曲和延伸链构件,并散布于整个蛋白;在N-端含有一个跨膜区,包含两个保守结构域,位于N-端的Pfam DUF3357 结构域和Glyco_32 结构域,属于糖基水解酶基因家族32超家族;系统进化表明与龙眼酸性转化酶基因同源,不同组织间LcSAI 表达水平为雄花>根>嫩茎>种子>嫩叶>雌花>果皮>老叶,果实不同发育阶段LcSAI 表达具有特异性。本研究为深入研究LcSAI 果实酸性转化酶基因调控蔗糖代谢途径机理提供数据依据。

  • XTH基因家族在妃子笑荔枝花穗不同处理方式的表达分析

    Abstract: Xyloglucan endotransglucosylase/hydrolase (XTH) is an important class of glycoside hydrolase that plays an important role in promoting plant cell wall elongation and responding to stress. In order to study the expression of the XTH family genes in different spike treatments, this study used the Feizixiao litchi as the experimental material. The flower spikes of litchi were treated with sparse flowers and uniconazole treatment respectively. Based on the pre-transcriptome database, 29 LcXTH family members were identified. The expression of LcXTH gene family members in different spikes was studied by real-time PCR (qRT-PCR). And analyzed the response of LcXTH family genes to different spike treatments. The results showed that the expression of LcXTH family members was different in the different developmental stages of the flowering stage after control, sparse treatment and uniconazole treatment, And there are differences in the expression levels of different genes. Among them, members of LcXTH subfamily III, LcXTH subfamily IV (LcXTH2, LcXTH3, LcXTH4, LcXTH5, LcXTH6, LcXTH7, LcXTH9, LcXTH10, LcXTH11, LcXTH12, LcXTH13, LcXTH22 and LcXTH23) and LcXTH20 have higher expression levels, presumably it plays a key role in flower development. LcXTH expression was up-regulated at the 14th day after flowering, We speculatee flowering has a positive regulation effect on LcXTH. Uniconazole treatment inhibited the expression of LcXTH and negatively regulated LcXTH.
